The official Kayako helpdesk works at around that scale, and some of our customers are actually much larger than that.

Sorry, hit post earlier than I'd meant to. As long as you're using a scalable webserver (Apache, Nginx, lighttpd, et cetera) and a relatively modern OS, you should be fine. Personally, I prefer CentOS, because it's rock solid stable, but that's an individual opinion. For a deployment of that size, you may be better off with a dedicated server. It's not required, but if you have that many people relying on a single tool, it probably shouldn't be subject to load from third parties you don't even know. -------------------------------------------------------------------

I would recommend CentOS and possibly looking into LiteSpeed as the web server.
