What do i (administrator) have to do so clients can respond to tickets via mail instead of going to the desk?
Now they only can reply via desk

You need to set up e-mail addresses and e-mail queues. This process is thoroughly documented in the manual: http://docs.kayako.net

Look into the "Setting up e-mail piping" section of the manual, that is what you need to do.
