Is your helpdesk installed on the main domain i.e. domain.com or a subdomain - subdomain.domain.com?

Check that the PHP path is correct in the /cli/index.php file for your server.
Code:
#!/usr/local/bin/php -q
Your server environment may be different.

What do you get when you run the file directly in your browser?
domain.com/esupport/cli/index.php
